Thermo Scientific Richard-Allan Scientific Neg 50 Frozen Section Medium

Instructions for Use

For in vitro diagnostic use.

For use as a frozen specimen embedding medium.

1. Cover the surface of the specimen holder with a thick layer of Thermo Scientific ™ Richard-Allan Scientific ™

Neg 50 ™ embedding medium. Use one color for each patient sample, or for each quadrant of a divided specimen.

2. Position and orient tissue on holder and press lightly to eliminate air bubbles beneath the tissue.

3. Apply additional Neg 50 embedding medium until the tissue is surrounded.

4. Freeze tissue to holder in usual manner until tissue and medium are firm. The color of the medium will lighten considerably but will still be easily identifiable.

5. Section in usual manner. If desired, place sections on microscope slides of the color matching the medium used for that specimen.

6. No special procedure is required to remove Neg 50 embedding medium from sections; it will dissolve in alcohol or aqueous solutions during the staining procedure.

7. To remove specimen from holder for further processing, dissolve medium and immediately place specimen in fixative.

8. To avoid vacuole formation within the cells (sometimes referred to as “freeze artifact”) it is best to freeze tissues as rapidly as possible. Tissues allowed to freeze slowly have sufficient time for the formation of ice crystals within the cells. These ice crystals expand inside the cells. When the ice crystals melt during staining, large holes or vacuoles remain inside the cells.
