Read these instructions carefully before using the products.
Intended use
The products are intended for research use only, and shall not be used in any clinical or in
vitro procedures for diagnostic purposes.
Safety
For use and handling of the products in a safe way, refer to the Safety Data Sheets.
Storage
Store at ambient temperature to expiry date. Once opened, completely reseal the pouch
containing the Ready-To-Go™ GenomiPhi™ HY DNA amplification kit cakes. To ensure
maximum shelf life and optimal performance storage in a desiccator is recommended.
Open product may be stored by resealing the pouch and folding the sealed edge over
several times and sealing with a clip. Store reconstituted Control DNA at -20°C. For
longer-term stability, once opened, store 2× Denaturation Buffer at 2–8°C.
DO NOT allow reconstituted reagents to warm above 4°C prior to amplification

Description
illustra™ Ready-To-Go GenomiPhi V3 DNA Amplification Kit is part of the Phi29 DNA
polymerase family of products from Cytiva. It contains all of the components necessary
for whole genome amplification by isothermal multiple strand displacement. The
reaction mixture is provided as predispensed, single-dose, lyophilized cakes in either
strips of 8 tubes, or 96-well plates.
Amplification is highly uniform over the entire genome so that locus representation
remains extremely close to the original DNA sample. Furthermore, amplification is
carried out with very high fidelity due to Phi29 DNA polymerase proofreading activity.
The average product length is over 10 kb

Note:
The Short Protocol is appropriate for researchers who have optimized their
GenomiPhi reactions. First-time users must review the Full Protocol. The full
protocol can be obtained by visiting cytiva.com or by contacting your local
Cytiva Technical Support Group.

Action

Heat denaturation of template in sample buffer
Mix 1 μl of template DNA (at least 10 ng) with 9 μl of Hâ‚‚0 and 10 μl 2×
denaturation buffer
Heat to 95°C for 3 min.
Cool to 4°C on ice.

Preparation of amplification reaction
For each amplification reaction add the 20 μl denatured sample to the ReadyTo-Go GenomiPhi V3 freeze-dried cake.

Incubation at 30°C
Close tubes with caps (provided) and incubate the sample at 30°C for 90
minutes (2 hours suggested for crude lysates).

Post-amplification heat inactivation
Heat the sample to 65°C for 10 min.
Cool to 4°C.

Optional purification of amplified products
Purify amplified material if necessary.
