1. Title of the procedure: Use of the autoclave
2. Hazard Assessment :

Autoclaves use superheated steam to sterilize materials. The high temperature (120-135 C) and
pressure (15 psi (103 kPa)) have potential to create burning or explosion
3. Precautions and Personal Protection Equipment :









Protective equipment should be worn at all times including gloves and lab coat
If the autoclave room fills with steam, press the red button on the wall and evacuate the room.
If this is not possible, evacuate the room, close the door, and press the red button outside the
room. Notify reception/lab technician
Leave room in containers for expansion. Do not overfill containers containing liquids
Ensure bottle caps are loosened before placing bottles in autoclave
Load the basket with the weight evenly distributed
Always check the pressure gauge to make sure that it is reading 0 psi before opening the door
Check the basket contents to ensure it is safe to remove e.g. check for breakages or leaks
Beware of liquid dropping from the basket. Take care when lifting bottles as lids will be loose
If the machine is not operating properly, notify the lab supervisor. Do not attempt repairs
4. Decontamination Procedures
Clean and wipe the work area and instrument with 70% ethanol if they are contaminated
(including the seal)
5. Specific Procedures












Use only Type I borosilicate glass bottles. Do not use ordinary glass bottles or any container
not designated for autoclave use, including plastic
Use the wire basket for most equipment. Use the solid basket for media and most waste. Load
the baskets with the weight distributed evenly
Leave space in between items for steam penetration.
Place the temperature probe in the densest part, and do not close the lid on it
Place an indicator strip in the basket
Cycles to be used: cycle 2: 122°C for 30 min (tips and equipment)
cycle 3: 135°C for 60 min (waste)
Close the door and select the cycle then press start
o
The door will only open when the temperature is <50 C. To open the door, press the ‘door’
button, display will read ‘hold’, wait 20 seconds, press door button again, move leaver to the
right to open
Check the indicator strip has passed. The blue tick should have turned black
Use the drying oven to dry pipette tips
Keep non-autoclaved and autoclaved items in designated areas
Fill in the log sheet. Check the print out indicates the temperature record is ok, sign the log
sheet and store the print out
