QIAGEN Maxi, Midi exoRNeasy Serum/Plasma Kits Quick-Start Protocol

exoRNeasy Serum/Plasma Kits Maxi and Midi are designed for isolating total RNA, including small RNAs, from exosomes and other EVs. The protocol begins with a vesicle isolation step and continues with RNA isolation, using the exoRNeasy Serum/Plasma Maxi and Midi Kits. The protocol provides detailed instructions for each step, including sample preparation, lysis, phase separation, and RNA elution.

Frequently asked questions

The exoRNeasy Serum/Plasma Maxi and Midi Kits are shipped at ambient temperature. Store the RNeasy® MinElute® spin columns immediately at 2–8°C. Store the miScript® Primer Assay at –15°C to –30°C. QIAzol® Lysis Reagent can be stored at room temperature (15–25°C) or at 2–8°C. Store the remaining components dry at room temperature.

The miRNeasy Serum/Plasma Spike-In Control (cat. no. 219610) is used to monitor the efficiency of the RNA isolation process. It is added to the sample before lysis. For recommendations on how to prepare a working solution, see the exoRNeasy Serum/Plasma Handbook.

After the final wash step, place the RNeasy MinElute spin column in a new 1.5 ml collection tube (supplied). Add 14 µl RNase-free water directly to the center of the spin column membrane. Close the lid gently, let the column stand for 1 min and then centrifuge for 1 min at full speed to elute the RNA.
