ÄKTA start UV Module and Support Information
Introduction
When ÄKTA start is installed for the first time, and when the UV flow cell has dried out
due to long storage, the following error and warnings can occur:
•
Error 101
•
Warning 115
•
Warning 116
Action: The UV flow cell must be refilled with demineralized water and the UV flow cell
calibration and baseline must be checked. The instructions below describe the steps
that shall be taken to correct the error and warnings listed above.
Prerequisites
The following prerequisites must be fulfilled before the corrections start:
•
Make sure that the UV flow cell is assembled tightly.
•
Flush the UV flow cell with demineralized water using the pump following the instructions below. Make sure that there are no air bubbles in the UV flow cell.
Flush the UV flow cell
Perform a wash of the UV cell by using the Washout fractionation tubing procedure,
which is initiated from the instrument display.

Immerse the buffer A inlet tubing in demineralized water.

In the ÄKTA start home screen, select Method run.

In the Method run screen, select Prepare system.

Select Washout fractionation tubing, and then tap Select.
In the Run parameters screen, set run parameters.
Select Run to initiate the wash.

Result: The Frac tube wash in progress screen opens.

When the run is ended, select Exit to close.
Check UV status
When the UV flow cell is flushed with demineralized water, check the UV status. Follow
the instructions below.

In the ÄKTA start home screen, select Settings and service, then select the
Settings and service screen 3 by tapping Next.

In the Settings and service screen 3, select UV.

In the UV screen, select UV LED calibration.

In the UV LED calibration screen, read the signal.
The UV LED intensity shall have a minimum signal of 2700 mV.
If the signal is < 2700 mV, follow the instructions below.
If the signal is > 2700 mV, refer to Set baseline (optional), on page 5.
UV LED calibration
Follow the instructions below to perform a calibration if the UV signal is below 2700 mV.

In the UV LED calibration screen, select Calibrate.
Result: A confirmation screen opens after calibration is complete.

Tap OK in the confirmation screen.

In the UV LED calibration screen, tap Save to store the calibrated Light
Strength value.
If the value is not saved, the same error messages will appear during next restart of the instrument.
Result: The system is ready to use.
Note:
An alternative is to use the up/down arrows, or type in the value, to increase
the Light Strength until the signal is above 2700 mV.
Set baseline (optional)
Follow the instructions below to set baseline if the signal is above 2700 mV.

During power up,

reaching the detector, i.e., too high absorbance in the cell
or too weak light
source.

Clean UV flow cell.

DM water.

Warning 115:
Flush flow cell
and mount securely
Flow cell not mounted properly or is optically misaligned.
Mount the flow cell properly. Calibrate the
UV module with flow cell filled with demineralized water.
Warning 116:
UV base lining
ignored
Flow cell is not
clean.
Clean the flow cell thoroughly with DM water. Calibrate the UV module with the flow
cell filled with demineralized water.
