Evolve™ Process Columns
Introduction
This document enables the integration and installation of a Bubble Trap to the Evolve™ Process Columns.
Bubble traps remove air from the system protecting the column from air ingress which can lead to the drying out
of the chromatography media/adsorbent, loss of packed bed integrity and loss of chromatographic performance.
Bubble traps also act as pulse dampeners, provide an equal and smooth flow which prevents excessive sample
dilution and poor flow distribution.
There are three bubble trap kits available - AABTK1 for 70 and 100 mm Evolve™ Process Columns (3 mm id
tubing), AABTK2 for 200 and 250 mm Evolve™ Process Columns (6 mm id tubing) and AABTK3 for 140 mm
Evolve™ Process Columns (6 mm id tubing).

Fitting Instructions
General Guidelines
To disassemble and assemble the bubble trap follow the instructions below, attention should be paid to:
1. Always use the correct size tools. A comprehensive tool kit is provided with the Evolve™ Process Column Kit.
2. Do not over-tighten components and use correct torque settings as given in this original Evolve™ Process
Column Kit operating manual.
3. Good engineering practices should be followed at all times and operations carried out in an approved and safe
manner.
Installation of Bubble Trap Kit
1. Assemble the side mounted accessory support bracket (i.e. larger diameter hole) to the bubble trap around the
centre of the acrylic tube. Please note: ensure that the bolts are tightened so the bubble trap is secure.
2. Attach the assembled side mounted accessory support bracket (i.e. smaller diameter hole) and bubble trap
around one of the stainless steel tie bars on the side of the Evolve™ Process Column. Please note: ensure that
the bolts are tightened securely.
3. Connect the T-Piece connector and manual pinch valve to the top of the bubble trap, using the ladish seals and
click clamps provided (see Figures 1 and 2 for reference). Please note: secure a blanking cap using a ladish seal
and click clamp to the open end of the T-Piece connector.
4. Connect the 4 Port 2 Way valve to the side outlet of the bubble trap, using the ladish seals and click clamps
provided (see Figures 1 and 2 for reference).
5. Connect the longer length of pipework (flexible hose) to the top port of the 4 Port 2 Way valve, using the ladish
seals and click clamps provided (see Figures 1 and 2 for reference). Please note: this is to be attached to the top
of the Evolve™ Process Column post priming.
6. Connect the shorter length of pipework to the bottom port of the 4 Port 2 Way valve and the bottom of the
bubble trap, using the ladish seals and click clamps provided (see Figures 1 and 2 for reference).
7. The flow from the pump or liquid chromatography system is connected to remaining port of the 4 Port 2 Way
valve (i.e. Process In – see Figures 3 and 4).

Operating Instructions
Bubble Trap Priming
1. After installation, the bubble trap and pipework must be primed before use.
2. With the bubble trap bypassed, if already attached, disconnect the pipework from the top of the Evolve™
Process Column (see Figure 4 for reference).
3. Turn on the pump and prime the pipework.
4. When a steady flow of buffer or solvent is achieved, switch of the pump and fit a blanking cap to the
pipework.
5. Switch the 4 Port 2 Way Valve to ensure the bubble trap is online (see Figure 3 for reference) and open the
manual pinch valve at the top of the bubble trap.
6. Turn on the pump and the bubble trap will start to fill.
7. When the bubble trap is approximately 2/3 full, turn off the pump and close the manual pinch valve.
8. Remove the blanking cap from the pipework and re-connect the hose to the top of the Evolve™ Process
Column.
9. The bubble trap is now ready to use.
Sample loading
To prevent over dilution or mixing effects while the sample is being applied to the column, the bubble trap
should be temporarily isolated from the system.
1. Turn the 4 Port 2 Way Valve handle to bypass the bubble trap and direct the flow straight to the column
(see Figure 4 for reference).
2. Once the sample application is complete return the 4 Port 2 Way Valve handle to its original position
(online, see Figure 3 for reference) to redirect flow through the bubble trap.

Cleaning and Storage
Cleaning
Please Note: Do not use abrasive cleaning materials or pads, as these may score plastic components.
The extensive solvent and chemical resistance of the bubble trap components allows the use of a wide range of Clean
in Place (CIP) solutions, including:
•
Sodium hydroxide (0.5 M - 1.0 M)
•
Sodium hydroxide (0.1 M) + ethanol (20%)
•
Peracetic acid (300 ppm)
For thorough cleaning after use the column should be dismantled into its component parts and thoroughly washed
with detergent and rinsed with RO water. The column should then be dried prior to storage.
Storage
Empty bubble traps should be kept in a cool, dry, dust-free location and protected from light. In particular, do not
expose acrylic column tubes to direct sunlight for prolonged periods or sterilize with ultra-violet lights as this may
reduce the mechanical strength of the tube.
