The maximum amount of starting material depends on the type of tissue and its RNA content. Generally, you can process up to 100 mg of brain or adipose tissue per RNeasy Mini spin column. For other tissues, a maximum of 50 mg tissue can be used, but for tissues with high RNA content like liver, spleen, and thymus, it is recommended to use no more than 30 mg.

You can disrupt and homogenize the tissue sample using either the TissueRuptor (for individual samples) or a TissueLyser system (for multiple samples simultaneously). The TissueRuptor uses a rotor-stator homogenizer to disrupt and homogenize the sample in QIAzol Lysis Reagent, while the TissueLyser systems use bead-milling, where the tissue is disrupted by rapid agitation in the presence of beads and lysis buffer.

The RNeasy Microarray Tissue Mini Kit purifies all RNA molecules longer than 200 nucleotides, making it particularly suitable for microarrays. This results in an enrichment for mRNA, as most RNAs smaller than 200 nucleotides are excluded.
