2 Product Description
MBT Galaxy HCCA Matrix IVD (referred to as 'Galaxy HCCA'; HCCA = α-Cyano-4-hydroxycinnamic acid) enables easy and convenient preparation of MALDI matrix solution for MALDI- TOF- MS measurement of peptides and proteins. Each vial contains 7.5 ±0.3 mg matrix, enabling fast preparation of the desired concentration by adding the relevant volume of solvent.

3 Inspection, Storage and Stability
3.1
Inspection on Arrival
Check the Galaxy HCCA package on arrival. If it is damaged, check the vials. If the vials are damaged, the Galaxy HCCA must not be used. Dispose of the Galaxy HCCA (vials and package) following the guidelines outlined in the Material Safety Data Sheet for the product and contact Bruker Daltonik
GmbH Service Department ([email protected]) for a replacement.

Storage on Arrival
+8°C Galaxy HCCA is shipped at ambient temperature.
The expiry date on the package is valid for the product when stored in a refrigerator at 2°C to 8°C on arrival.

5 Field of Application
Galaxy HCCA enables easy and convenient preparation of HCCA matrix solutions using the MBT
Galaxy. The matrix is soluble in standard solvent (see section 6), is easy to handle, and enables highly
sensitive MALDI-TOF-MS measurement of peptides and proteins from 0.7 to 20 kDa.
MALDI-TOF-MS using HCCA as a matrix has been shown to be a reliable, high-throughput method for the classification and identification of microorganisms.

6 Solubilization of Galaxy HCCA
Chemicals and Materials Required

Standard solvent (acetonitrile 50%, water 47.5% and trifluoroacetic acid 2.5%) from Sigma-
Aldrich (# 19182) or OS solution from VWR (# PRLS89449.230), which have been tested by
Bruker Daltonik GmbH and are recommended for reconstitution of Galaxy HCCA.

Solubilization Procedure
The procedure for solubilization of Galaxy HCCA depends on the type of MALDI target plate to be used.
For BigAnchor targets
1. Add 1.5 mL standard solvent to the screw-cap vial containing Galaxy HCCA (final concentration:
5 mg Galaxy HCCA/mL) and close the vial tightly.
2. Shake the screw-cap vial and use a vortex mixer to completely dissolve Galaxy HCCA at room temperature. Finally, shake the contents down and tap the closed vial on a tabletop so that any liquid that has collected inside the screw cap falls into the vial.
3. Remove the screw cap and exchange the matrix vial in Channel 1 of the MBT Galaxy liquids drawer as described in the MBT Galaxy User Manual.
For all other targets approved for your workflow
1. Add 750 µL standard solvent to the screw-cap vial containing Galaxy HCCA (final concentration:
10 mg Galaxy HCCA/mL) and close the vial tightly.
2. Shake the screw-cap vial and use a vortex mixer to completely dissolve Galaxy HCCA at room temperature. Finally, shake the contents down and tap the closed vial on a tabletop so that any liquid that has collected inside the screw cap falls into the vial.
3. Remove the screw cap and exchange the matrix vial in Channel 1 of the MBT Galaxy liquids drawer as described in the MBT Galaxy User Manual.

7 Sample Preparation Using Galaxy HCCA
The procedures below can be used for the classification and identification of microorganisms through
MALDI-TOF-MS.
CAUTION If Galaxy HCCA matrix solution is not added to samples within 30 minutes after they have dried, these positions cannot be tested.
Direct transfer sample preparation procedure
1. For each sample, smear an isolated colony as a thin film directly onto a sample position using a sterile wooden stick.
2. Deposit 1 μL of IVD BTS onto at least one MALDI target plate position.
3. Allow the sample spots to dry at room temperature.
4. Perform the matrix only MBT Galaxy preparation workflow as described in the MBT Galaxy User
Manual.
5. Perform MALDI-TOF-MS measurement.
extended Direct Transfer sample preparation procedure
1. For each sample, smear an isolated colony as a thin film directly onto a sample position using a sterile wooden stick.
2. Deposit 1 μL of IVD BTS onto at least one MALDI target plate position.
3. Allow the sample spots to dry at room temperature.
4. Perform the formic acid + matrix MBT Galaxy preparation workflow as described in the MBT
Galaxy User Manual.
5. Perform MALDI-TOF-MS measurement.
Extraction sample preparation procedure
1. Follow the extraction sample preparation procedure up to the point where the supernatant is deposited onto the MALDI target plate.
2. Deposit 1 μL of IVD BTS onto at least one MALDI target plate position.
3. Allow the sample spots to dry at room temperature.
4. Perform the matrix only MBT Galaxy preparation workflow as described in the MBT Galaxy User
Manual.
5. Perform MALDI-TOF-MS measurement.
