Frequently Answers and Questions

How do I replace the mercury burner?

Read chapter 9, "ASSEMBLY", in this instruction manual for details on replacing the mercury burner.

What safety precautions should I take when using the microscope?

Make sure that the mercury burner is attached and cords are connected. The internal parts become very hot during and within 10 minutes after completion of operation. Do not attempt to open the light housing in these periods. Always use the power cord provided by Olympus. Ensure that the grounding terminal of the microscope and that of the wall outlet are properly connected.

How do I select the appropriate fluorescence mirror unit for my observation?

Select the fluorescence mirror unit that provides excitation matching the fluorochrome you are using. Consult the instruction manual for details on the available mirror units and their configurations.

How do I center the mercury burner?

Follow the instructions on pages 17-18 of this instruction manual. The burner should be centered so that the arc image is overlaid with the mirror arc image.

Can I use the reflected fluorescence system with other observation methods?

Yes, you can use the system with transmitted light brightfield observation, transmitted light phase contrast observation, and transmitted light DIC (Differential Interference Contrast) observation. You can also perform reflected fluorescence observation simultaneously with phase contrast or DIC observation.

How to install modules in the IX2-RFAC?

To install modules in the IX2-RFAC, follow these steps:

  1. Remove the objective unit replacement cover: Remove the magnet attached by magnetism.
  2. Remove the dummy objective unit: Remove the dummy unit from the position where you want to install the fluorescence objective unit by loosening the clamping screw of each dummy objective unit and moving it outward.
  3. Install the fluorescence objective unit: Insert the fluorescence objective unit into the mounting dovetail so that the model indication of the objective unit faces down, push it in completely, and securely tighten the clamping screw.
  4. Insert the remaining fluorescence objective units: Be sure to leave the dummy objective unit in the position where the fluorescence objective unit is not installed.
  5. Insert the objective unit indication plate: Insert the objective unit indication plate into the indication plate of the microscope frame so that each indication plate matches the turret position number of the corresponding fluorescence objective unit.
  6. Install the cover: Return the cover to its original position.

How to view fluorescence images?

Fluorescence image viewing requires the IX2-UCB control box, U-HSTR2 manual switch, a computer, and IX2-BSW software for motorized component control. A computer connection is necessary.

Where to place the sample for fluorescence microscopy?

Place the sample on the specimen stage before fluorescence observation.

How to connect a mercury lamp to a microscope?

Loosen the two fixing screws (1) on the lamp housing mounting holes using a hexagon wrench. Insert the lamp housing completely. Tighten the screws. Ensure sufficient space around the lamp, especially above, as it gets hot. Do not install at an angle. If using the U-FWR filter wheel, connect modules from reflected light illuminator - IX2-SHA motorized shutter - U-FWR filter wheel - lamp housing.

How to achieve fluorescence observation after turning on the mercury lamp?

  1. Prepare: Install the fluorescence unit and appropriate objective lens. Center the mercury lamp filament. Optionally use the 32FR80 frost filter for even illumination (reduces intensity by 20%). If using IX2-RFACA or motorized shutter, turn the main switch 1 on the IX2-UCB control box to ON. 2. Turn on the main switch to "I" (ON) and wait 5-10 minutes. 3. Place the sample. 4. Select the unit. 5. Focus. 6. Open the shutter. 7. Adjust light intensity with ND filter (if needed). 8. Adjust the condenser. 9. Adjust field diaphragm. 10. Adjust aperture diaphragm (IX2-RFAL L-shaped reflected light illuminator only). 11. Observe. Close the shutter if interrupting observation.

How to connect the mercury lamp to the microscope?

To connect the mercury lamp, use a hex wrench to loosen the two fixing screws of the lamp socket installation hole. Fully insert the lamp socket and tighten the two fixing screws. Ensure sufficient space around the lamp housing, especially above the lamp socket, as it becomes very hot during operation. If using a U-FWR filter wheel, connect the modules in the order: Reflected light illuminator - IX2-SHA motorized shutter - U-FWR filter wheel - Lamp housing.

What is the procedure for observing fluorescence after turning on the power?

After turning on the main switch, wait 5 to 10 minutes for the arc image to stabilize. Install the appropriate fluorescence mirror unit and objective for the observation method. Place the sample on the stage, select the mirror unit, and focus the objective. Open the shutter and adjust the light intensity using ND filters. Finally, adjust the condenser, field iris diaphragm, and aperture iris diaphragm for uniform illumination.

Is it possible to use the IX2-RFACA system to generate fluorescence images without an external computer?

No, the IX2-RFACA motorized fluorescence mirror unit cassette requires an IX2-UCB control box and a PC with IX2-BSW software to control the motorized unit switching. Additionally, the descriptions for the IX2-UCB and U-HSTR2 are in a separate manual. Therefore, it is not possible to generate images without connecting to a computer.

How to connect the mercury lamp to the microscope?

Use a hexagon wrench to loosen the two fixing screws of the lamp holder socket. Insert the lamp holder completely and tighten the fixing screws. Ensure sufficient space around the lamp holder as it becomes very hot; do not install it in a tilted position. If using a U-FWR filter wheel, connect the modules in the following order: Reflected light illuminator - IX2-SHA motorized shutter - U-FWR filter wheel - Lamp holder.

How to observe fluorescence after turning on the mercury lamp?

  1. Preparation: Install the fluorescence mirror unit and objective suitable for the observation method, and center the mercury burner filament. 2. Turn on the power: Turn the main switch to 'I' (ON) and wait for the arc image to stabilize (5 to 10 minutes). 3. Place the specimen on the stage. 4. Select the mirror unit matching the specimen. 5. Focus: Place the objective in the light path and focus on the specimen. 6. Open the shutter. 7. Adjust light intensity: Insert ND filters into the light path as needed. 8. Adjust the condenser for even and brightest field of view. 9. Adjust the field iris diaphragm. 10. Adjust the aperture iris diaphragm (IX2-RFAL only). 11. Start observation.
