Frequently asked questions

To start up, uncover the microscope, tilt the illumination arm back, move objectives down, turn on the Hg-lamp (if needed), laser power switch and key, Scan box, and computer/microscope. Log in via the iCenter application.

A good starting laser power for the Argon laser is 20%.

First, turn off the key switch for lasers (leaving the power switch on for cooling), clean objectives, transfer data to the server, and sign out. Then shut down the computer (if applicable), scan head, and Hg lamp, finally covering the scope after the laser cooling fan stops.

Use high gain and low laser power (starting gain around 1000). Check for bleed-through by turning down each laser. Use the quick-lookup table (rainbow icon) to identify maxed-out (blue) or black (green) pixels and adjust for a good dynamic range.
