Setting up the Nikon TE2000-E microscope for DIC Imaging
1. Turn the microscope power (A) on at
the back of the scope

2. Turn the bright field light source on
3. Make sure the DIA lamp is ON on the
microscope controller box (B)
4. Make sure the light path is directed to
they eyepieces on the controller box (B)
5. Light intensity can be adjusted with

up/down buttons labeled ADJ on
controller box (B)

6. Put a slide on the stage and focus it

with the lowest power objective you will
F
use (note: not all low power objectives
are DIC – it’s written on the nosepiece)
7. Close down the field diaphragm (C) all
the way. You should see a bright spot
surrounded by darkness.
8. Close down the condenser aperture (E)
by pushing the lever to the left
9. Focus the edges of the spot with the
condenser focus knobs (D). You want a
A
sharp octagon with a slightly blue halo
10. Center the Octagon in the field of view
11. Open the field diaphragm until the light
just fills the field of view
12. Adjust the condenser aperture
diaphragm (E) to about 80% open. If
you start to see a glare from your
B
sample, you’ve opened it too far.
13. Rotate condenser (F) to DIC N1 (for
10x objective) or N2 (for 20x -100x)
14. Insert polarizer (G) in light path
15. Make sure analyzer is IN on controller
box (B)
16. For better contrast you can play with the rotation of the polarizer (G) by loosening
the screw on top and turning it by holding the silver handle.
17. You will need to adjust the field diaphragm and condenser aperture if you change
objectives.
To capture images using elements:
1. Turn on the computer
2. Open Elements
3. Select “Roper Scientific Camera” in the dialog box
4. Redirect the light path to the camera (down) on the microscope controller box or
in the TE2000 control pad in elements
5. Select Image Setup “DIC 20x – 100x” from the main menu bar
6. Hit the “live” button, auto expose (AE) and capture the image (see separate
sheet for more details about using Elements.
